ABSTRACT: Puerariae radix, the dried root of Pueraria lobata (Willd.) Ohwi, is one of the most important edible crude herbs used for various medical purposes. Pueraria lobata flavone, a kind of major active components, has been confirmed to play a remarkable curative eect on cerebral ischemia, but the mechanism is not clear. Therefore, in this study, 1H-NMR-based metabonomics approach has been used to investigate the protective eect of PLF on rats of cerebral ischemiareperfusion. Through principal component analysis, it was observed that metabolic alterations induced by ischemic cerebral-reperfusion were restored after treatment with PLF. Metabolites with significant changes induced by ischemic cerebral-reperfusion, including lactate, taurine, glutamate, choline, glucose, lipids, creatine, which were characterized as potential biomarkers involved in the pathogenesis of ischemic cerebral-reperfusion. All those biomarkers can be regulated by Pueraria lobata flavone treatment, which suggested that the therapeutic eect of PLF on ischemic cerebral-reperfusion may involve in regulating the dysfunctions of energy metabolism, amino acid metabolism, energy metabolism. This study indicated that the Pueraria lobata flavone had protective eect on rats of cerebral ischemia-reperfusion and explored the metabolic regulation mechanism and the NMR-based metabonomics approach might be a promising approach to study mechanisms of traditional Chinese medicines.
