ABSTRACT

A designer nuclease is a nuclease engineered to cleave a very specific phosphodiester sequence in a duplex DNA (double-strand DNA cleavage). Current designer nucleases include meganucleases, zinc finger nucleases (ZFNs), transcription activator-like effector nucleases (TALENs), and clustered regulatory interspaced short palindromic repeat (CRISPR)-associated protein 9 (CRISPR/Cas9). TALENs and ZFNs contain a restriction nuclease, Fokl. Meganucleases and CRISPR/Cas9 contains an intrinsic nuclease function.
