ABSTRACT

A versatile toolbox tailored for opsin-free optogenetics, including allosteric switches, photo-inducible multimerization and dissociable systems, has been developed and achieved remarkable success in the precise control of cell physiology and host behaviors with high spatial and temporal precision. This chapter focuses on the integration of optogenetics with store-operated calcium entry (SOCE) to illustrate the use of optogenetic tools to delineate the activation and deactivation kinetics of signaling activities, study subcellular protein-protein interactions (PPIs) at high spatiotemporal resolution, and probe protein conformation changes and oligomerization.
