ABSTRACT

This chapter provides information on drug combination screening. Sulfinpyrazone was reported to interfere with the acetylsalicylic acid effects on cyclo-oxygenase activity ex vivo in rats probably by the competition with the binding site. The study introduced another factor which may be of importance for drug combinations: the sequential principle. Thus, the inhibitory adenosine effects on white body formation in the rabbit and rat cerebral cortex microcirculation were particularly potentiated by dipyridamole and potentiation was likewise observed in vitro by Dawicki et al., who also used some drugs related to dipyridamole. Some interesting observations were also recorded with other agents such as the potentiation of heparin effect by some adrenergic blocking agents, potentiation of ketanserin by dipyridamole, and possibly by troxerutin and pentoxifylline.
