ABSTRACT

Amorolfine is a morpholine antifungal agent for topical use. It is commonly available in the form of a nail lacquer, containing 5% amorolfine as the active ingredient to treat onychomycosis. Amorolfine appears to have a low allergenic potential. However, amorolfine nail lacquers could have higher allergenicity as they contain excipients such as butyl acetate, ethyl acetate and ethanol that evaporate after the application of the product, thereby increasing the concentration of amorolfine in the nail plate and in the perionychium. Patch tests were positive to the nail lacquer ‘as is’ and – later – to amorolfine 1.0% pet. Seven controls were negative. Additional patch tests were carried out with amorolfine and the other ingredients of the commercial cream and showed positive reactions to amorolfine 0.25% and 0.5% pet. Patch tests showed positive reactions to the pharmaceutical cream and – later – to amorolfine 0.3%, 0.5% and 1% pet. and 1% water.
