ABSTRACT
Once a bioactive lead has been identified, there is still a lengthy process involved in converting that lead to a drug used in humans. While there are notable exceptions where the initially discovered hit or lead becomes a drug, most modern drug discovery requires a process of lead optimization. Lead optimization involves chemical modification of the structure of a hit or lead compound through an iterative series of design, synthesis, and testing.
