TheamplificationofDNAisoneofthebasictechniquescommontoallmolecularbiology laboratories.AmplificationisneededtoincreasethequantityoftheDNAofinterest.ThisDNA, intum,isputtouseastheneedsofeachlaboratorydictate.Amongthemyriadofuses,amplified DNAmaybeemployedsimplyasaprobe(i.e.,hybridizations),usedforfurthercharacterization oftheDNA(i.e.,sequencing,fingerprinting);ormaybeserveasatemplateinprotocolssuchas site-directedmutagenesisandDNAfootprinting.Mostofthischapterisdevotedtotheamplification ofDNA;addressingisothermalamplificationandthermalcyclingamplificationofDNA.Theend ofthischapterpresentsamethodofgeneisolationthatincorporatesathermalcyclingamplification procedure.
