ABSTRACT

Hybridization is carried out on tissue before it is embedded in resin. Semithin and ultrathin sections can then be observed with light and electron microscopes, respectively. This chapter discusses the parameters for cutting vibratome sections, storage of vibratome sections and principles of permeabilization, deproteinization, prehybridization and hybridization. It also discusses different types of epoxy resin. Fixation, embedding and visualization of vibratome sections are reviewed. The chapter also reviews radioactive hybrids, antigenic hybrids and visualization of ultrathin sections.
