ABSTRACT

Where Et (or some textbooks have it as Eo) represents starting/initial/total amount of enzyme.

The terms were collected in such a manner that k2 and k−1 appeared as the numerator and then the Km, the Michaelis-Menten constant, assigned to the fraction. Both k2 and k−1 represent the breakdown of ES. Hence, the greater the Km, the greater the instability of ES or lack of affinity of E for S; it does not mean a faster enzyme catalysed reaction (producing P).
