This chapter describes in detail the protocols for the construction and screening of a bacteriophage λ library.1-7

Traditionally, the widely used bacteriophage λ DNA vectors are EMBL3 and EMBL4 (Figure 15.1). Both types of vectors contain a left arm (20 kb), a right arm (9 kb), and a central stuffer (14 kb) that can be replaced with a foreign genomic DNA insert (9 to 23 kb). The difference between two λ DNA vectors is the orientation of the multiple cloning site (MCS).
