ABSTRACT

I. INTRODUCTION The methods for the derivatization of sugars described elsewhere in this volume are based on reductive amination of reducing sugars or their con­ densation with suitable pyrazolones. The preliminary account in this chap­ ter describes an alternative technique in which UV-absorbing derivatives are formed from glycosylhydrazones under mild conditions. The method could be particularly useful for the analysis of the products of hydrazinolysis of glycoproteins, because it avoids the hydrolysis of the glycan hydrazones and any associated incompleteness of recovery. This method can also be used for reducing sugars, because they can be converted quantitatively to the hydrazones by treatment with hydrazine hydrate at room temperature.
