In the Lowry assay, you record the extinction of a series of samples with known protein concentration, from which you construct a line of best t. en, the concentration of an unknown sample can be determined by nding the concentration corresponding to an extinction value on that

line. Back in 1951, the line of best t was drawn on paper (see Figure 7.1), and the unknown sample was determined manually. Most of this chapter is about handling a lot of sample data in Python using Lowry’s extinction values as an example. An example of a standard series was given by Lowry in Table IV (Measurement of Small Amount of Protein from Rabbit Brain) of his publication, where he determined the protein concentrations of 18 rabbit brain samples. He examined six concentrations three times each, obtaining the values reported in Table 7.1.
