Since the capillary is the initial barrier to distribution, its permeability characteristics will be our first concern. We shall then examine the general factors that influence the amount of drug eventually reaching its site of action. Finally, special attention will be directed to the singular aspects of passage of drugs into the brain and across the placenta into the fetus.
