ABSTRACT

Objectives: Determine the B.flabellifer L. fruit seed skin ethanol extract toxicity on HSC-3 cell line nuclear fragmentation and reduction. Method: negative, positive control (doxorubicin 3µM), and B.flabellifer L. seed skin extract groups with five concentrations applied to HSC-3 cells planted, incubated for 24-, 48-, 72-hours and observed after HE staining. Three of four fields of each group were analyzed, considering nuclear fragmentation present with at least one fragmented nucleus. Nucleus surface was measured in ImageJ for nuclear reduction assessment and analyzed by Chi-square and Kruskal-Wallis. Results: Nuclear fragmentation occurs in all extract groups at 24-, 48- and 72-hours, resulted in significant differences with the negative control (p=0.00<0.05). Significant differences with p<0.05 in nuclear reduction on 24-; 48-; 72-hours between the negative control and extract groups of 750 and 1500μg/mL; 375 and 750μg/mL; 375, 750 and 1500μg/mL respectively. Conclusion: B.flabellifer L. fruit seed skin has nuclear fragmentation and reduction effects.
