ABSTRACT
Objectives: This study aimed to evaluate the cytotoxic effects of AgNO3 synthesis with Cymbopogon Citratus (AgNPCc) on human dermal fibroblasts (HDF) using WST-8 tetrazolium salt [2-(2-methoxy-4-nitrophenyl)-3-(4-nitrophenyl)-5-(2,4-disulfophenyl)-2H-tetrazolium, monosodium salt] in CCK-8. Methods: Cytotoxicity tests were conducted using CCK-8 assay on six groups, consisting of one negative control and five concentrations (7.5 µg/mL, 15 µg/mL, 30 µg/mL, 60 µg/mL, and 120 µg/mL) of AgNPCc. After incubation with CCK-8 for 1 hour, WST-8 was reduced by dehydrogenase in cells to produce a yellow-colored product (formazan) soluble in the culture medium. The color change was measured at a wavelength of 450 nm. Results: Additionally, the IC50 value of AgNPCc was 78.79 µg/mL, showing the highest toxicity at a concentration of 120 µg/mL with more than 50% cell death. Conclusion: AgNPCc exhibited strong cytotoxicity against HDF at higher concentrations.
